Methodology, step by step…  to build a cellular model of AD, I stably transfected human hippocampal neurons with Swedish mutated gene APPSwe.  in order to determine the conditions of the experiments I conducted serious of dose-response assays using neutral red, which allowed me to determine the cytotoxicity of polyphenols studied in relation to control cells and transfected with Swedish mutated gene APPSwe. To accomplish main research aims I determined catalytic activity of SIRT1 and changes in its expression. SIRT1 is involved in the activation of multiple pathways, associated with neurodegenerative processes, thus I will assess:  level of amyloid β,  level of NEP (an enzyme involved in promoting degradation of amyloid beta-peptide),  level of BDNF (brain-derived neurotrophic factor, which is involved in mechanisms of synaptic plasticity in the adult brain)  level of NF-kB, which controls global pro-inflammatory response The activity of superoxide dismutase, catalase and glutathione reductase will be also determined. The examination of antioxidant properties of polyphenols will be based on assessment of chosen parameters for cells treated and untreated with oxidizing factor (ionizing radiation). The percentage of apoptotic and necrotic cells will be also estimated.
  • 4. Research project impact  Proposed comprehensive analysis will be useful in explaining the mechanism of protective influence of chosen compounds. The conducted experiments will make it possible to e.g. compare efficiency of polyphenols in neuronal cells different the amount of released Aβ,  Beside its purely cognitive aspects, it can have a substantial practical value in terms of the possible application. Introduction of compounds responsible for catalytic activity of sirtuin 1, will speed up therapies optimization due to application of relatively cheap and safety biofarmaceutics,  Gaining insight into mechanism of effects of resveratrol, epsilon-viniferin, piceatannol and xanthohumol on neuronal cells will also indicate the direction of research aiming at the development of the new strategies for prevention of age related diseases.
